Michael Wagner and Brian Thompson had two second-half scoring runs each and Seneca Wallace threw for 234 yards for the Cyclones (1-1, 1-0 Big 12), who outgained Kansas (0-1, 0-1) by 511 yards to 150.
The Iowa State defense, meanwhile, sacked Kansas' quarterbacks four times and set up three touchdown drives with interceptions.
Since falling behind No. 3 Florida State 24-0 in the second quarter of their season-opening 38-31 loss a week ago, the Cyclones have outscored the opposition 83-17.
Adam Benicke's 32-yard field goal on Iowa State's opening drive put the Cyclones up 3-0, but they had to punt when their next possession stalled at the Jayhawks' 35.
One play after that, though, Jeremy Loyd intercepted Zach Dyer's pass and returned it a yard to the Kansas 27. That set up Lane Danielsen's 9-yard TD run on an end-around, giving Iowa State a 10-0 lead with just under four minutes left in the first quarter and the rout was on.
Thompson, a redshirt freshman, finished with 96 yards on 14 carries, all in the fourth quarter.
Dyer was 6-for-14 for 50 yards and lost 16 yards on the ground before Mangino replaced him with junior college transfer Bill Whittemore to start the second half. Whittemore completed just 4-of-17 attempts for 31 yards and had one pass intercepted, but ran for 29 yards on seven carries and led the Jayhawks' only scoring drive.
